Does anyone know when the first 645 engine was built? The EMD 434 Demonstrator has a July 1964 build date, so I am thinking the first 645 engine would have predated this unit by six months or more.

It appears the first 645 was out in 1965. I have found that some dates vary depending on the site you visit. I would say it a safe bet 1965 and a year in either direction.

Since the demo came out in July 1964, I would say the 645 prime mover was probably developed in late 1963. It was basically a 567 with the 645 power assemblies installed. I am sure it probably took some time to get the bugs out before the demo went on the road.
